Gas Chromatography develops new applications for anaerobic activity testing

Most of them are anaerobic bacteria. Under normal circumstances, these bacteria are harmless to the human body, and even play an indispensable role in human growth and health. However, once the barrier function of the skin and mucous membranes is reduced or the bacterial flora is imbalanced, the anaerobic bacteria will cause infection and cause harm to health.
Anaerobic bacteria can infect almost all tissues and organs, causing oral infections, appendicitis, otitis media, osteomyelitis, sepsis, tetanus and many other diseases. Some diseases can also be life-threatening. The main treatment for anaerobic infections is broad-spectrum antibacterial therapy, which uses broad-spectrum antibiotics such as penicillin, amoxicillin, ofloxacin, etc. for treatment.
With the increasing resistance of anaerobic bacteria, the applicability of traditional broad-spectrum antibacterial therapy based on experience has begun to become lower and lower, and clinical treatment failures often occur. In order to solve this problem, it is necessary to establish a method for evaluating the efficacy of anti-anaerobic bacteria drugs based on tests and tests to accurately determine the anti-anaerobic activity of drugs. However, the cultivation of anaerobic bacteria requires a low-oxygen or anaerobic environment, and the growth of anaerobic bacteria is slow. Therefore, the anti-anaerobic test of drug candidates is often very costly and ineffective.
Recently, the research team of Wuhan Botanical Garden has developed a new method for determining the anti-anaerobic activity of candidate drugs by using headspace-gas chromatography (HS-GC-TCD). This method uses the characteristics of anaerobic bacteria to produce acid, and uses the metabolic acidity in the medium as a test indicator of anaerobic bacteria activity. The metabolic acid reacts with sodium bicarbonate to be completely converted into carbon dioxide, and then the content of carbon dioxide is detected by HS-GC-TCD. This method is more sensitive to cell activity, can greatly increase the accuracy of anti-anaerobic activity detection (relative standard deviation is less than 5%), and can reduce cross-contamination, suitable for routine screening of multiple drugs with anti-anaerobic activity.
Headspace-Gas Chromatography, also known as headspace gas chromatography, connects a headspace sampler to a gas chromatograph. The headspace sampler can specifically collect the volatile components in the sample or extract the adsorbed gas sample and then directly introduce it into the gas chromatograph for separation and detection, which is simple and practical. Headspace-gas chromatography has been widely used in environmental detection, organic residual solvent detection in food and pharmaceuticals. The new method developed by the research team not only provides an effective evaluation tool for anti-anaerobic drugs, but also opens up new application fields for headspace-gas chromatography.
